    Accuracy of adults’ recall of childhood social class: findings from the Aberdeen children of the 1950s study

    <b>Background</b>: Although adult reported childhood socioeconomic position has been related to health outcomes in many studies, little is known about the validity of such distantly recalled information. This study evaluated the validity of adults’ reports of childhood paternal social class. <b>Methods</b>: Data are drawn from the Aberdeen children of the 1950s study, a cohort of 12 150 people born in Aberdeen (Scotland) who took part in a school based survey in 1962. In this survey, two indices of early life socioeconomic position were collected: occupational social class at birth (abstracted from maternity records) and occupational social class in childhood (reported during the 1962 survey by the study participants). Between 2000 and 2003, a questionnaire was mailed to traced middle aged cohort members in which inquiries were made about their fathers’ occupation when they were aged 12 years. The level of agreement between these reports and prospectively collected data on occupational social class was assessed. <b>Results</b>: In total, 7183 (63.7%) persons responded to the mid-life questionnaire. Agreement was moderate between social class of father recalled in adulthood and that measured in early life ( statistics were 0.47 for social class measured at birth, and 0.56 for social class reported by the child). The relation of occupational social class to birth weight and childhood intelligence was in the expected directions, although weaker for adults’ reports in comparison with prospectively gathered data. <b>Conclusions</b>: In studies of adult disease aetiology, associations between childhood social class based on adult recall of parental occupation and health outcomes are likely to underestimate real effects

    Development of a passive compliant mechanism for measurement of micro/nano-scale planar three DOF motions

    This paper presents the design, optimization, and computational and experimental performance evaluations of a passively actuated, monolithic, compliant mechanism. The mechanism is designed to be mounted on or built into any precision positioning stage which produces three degree of freedom (DOF) planar motions. It transforms such movements into linear motions which can then be measured using laser interferometry based sensing and measurement techniques commonly used for translational axes. This methodology reduces the introduction of geometric errors into sensor measurements, and bypasses the need for increased complexity sensing systems. A computational technique is employed to optimize the mechanism’s performance, in particular to ensure the kinematic relationships match a set of desired relationships. Computational analysis is then employed to predict the performance of the mechanism throughout the workspace of a coupled positioning stage, and the errors are shown to vary linearly with the input position. This allows the errors to be corrected through calibration. A prototype is manufactured and experimentally tested, confirming the ability of the proposed mechanism to permit measurements of three DOF motions

    Laser-based sensing, measurement, and misalignment control of coupled Linear and angular motion for ultrahigh precision movement

    This paper presents a novel methodology for position and orientation (pose) measurement of stages used for micro/nano positioning which produce coupled motions with three planar degrees of freedom (DOF). In the proposed methodology, counter-rotation of the entire mechanism prevents the misalignment of the measurement beams within a laser-interferometry-based sensing and measurement technique. To detect such a misalignment, a sensing strategy constructed around a position sensitive diode has been developed. A feedforward-feedback compound controller has been established to provide the necessary counter-rotation input to reduce the misalignment error. Experimental validation has been conducted through the measurement of the workspace of a three-DOF planar micro/nano positioning stage. Experimental results demonstrate the capability of the technique to provide combined linear/angular measurement

    Enrichment of clinically relevant organisms in spontaneous preterm delivered placenta and reagent contamination across all clinical groups in a large UK pregnancy cohort.

    In this study differences in the placental microbiota of term and preterm deliveries from a large UK pregnancy cohort were studied using 16S targeted amplicon sequencing. The impact of contamination from DNA extraction, PCR reagents, as well as those from delivery itself were also examined. A total of 400 placental samples from 256 singleton pregnancies were analysed and differences investigated between spontaneous preterm, non-spontaneous preterm, and term delivered placenta. DNA from recently delivered placenta was extracted, and screening for bacterial DNA was carried out using targeted sequencing of the 16S rRNA gene on the Illumina MiSeq platform. Sequenced reads were analysed for presence of contaminating operational taxonomic units (OTUs) identified via sequencing of negative extraction and PCR blank samples. Differential abundance and between sample (beta) diversity metrics were then compared. A large proportion of the reads sequenced from the extracted placental samples mapped to OTUs that were also found in negative extractions. Striking differences in the composition of samples were also observed, according to whether the placenta was delivered abdominally or vaginally, providing strong circumstantial evidence for delivery contamination as an important contributor to observed microbial profiles. When OTU and genus level abundances were compared between the groups of interest, a number of organisms were enriched in the spontaneous preterm cohort, including organisms that have been previously associated with adverse pregnancy outcomes, specifically Mycoplasma spp., and Ureaplasma spp.. However, analyses of overall community structure did not reveal convincing evidence for the existence of a reproducible 'preterm placental microbiome'. IMPORTANCE: Preterm birth is associated with both psychological and physical disabilities and is the leading cause of infant morbidity and mortality worldwide. Infection is known to be an important cause of spontaneous preterm birth, and recent research has implicated variation in the 'placental microbiome' with preterm birth risk. Consistent with previous studies, the abundance of certain clinically relevant species differed between spontaneous preterm and non-spontaneous preterm or term delivered placenta. These results support the view that a proportion of spontaneous preterm births have an intra-uterine infection component. However, an additional observation from this study was that a substantial proportion of reads sequenced were contaminating reads, rather than DNA from endogenous, clinically relevant species. This observation warrants caution in the interpretation of sequencing output from such low biomass samples as the placenta
